Description
Oxoid™ D'Nase Agar (500g) is a differential medium used for the detection of deoxyribonuclease (DNase) activity in microorganisms, particularly Staphylococcus aureus. It is commonly used in clinical microbiology to distinguish DNase-positive pathogens from non-producers. The medium contains DNA as a substrate, and DNase-producing organisms hydrolyse the DNA, which can be visualised by flooding the plate with hydrochloric acid or toluidine blue, resulting in a clear zone around colonies.
Key Features:
- Form: Dehydrated powder
- Weight: 500g
- Application: Detection of DNase activity, primarily in the differentiation of Staphylococcus aureus and other DNase-positive organisms
- Composition: Contains DNA, peptones, and nutrients to support bacterial growth; designed to reveal DNA hydrolysis
- Preparation: Suspend 38g of the dehydrated medium in 1 litre of distilled water, mix thoroughly, and autoclave at 121°C for 15 minutes
- Yield: Approximately 13 litres of prepared medium
- Storage: Store the dehydrated medium at 10–30°C, protected from moisture and light
- Compliance: Suitable for use in clinical microbiology and research settings for DNase testing
